Use | Idarubicin HCl, a Idarubicin hydrochloride form, is an anthracycline antibiotic that inhibits DNA topoisomerase II (topo II) and acts on MCF-7 cells with an IC50 of 3.3 ng/mL. |
In vitro study | The IC50 of Idarubicin was 3.3±0.4 ng / mL in MCF-7 monolayers and 7.9±1.1 ng / mL in multicellular spheroids. In various in vitro systems, Idarubicin showed greater cytotoxicity than daunorubicin or doxorubicin. This is attributed to the fact that Idarubicin better induces the formation of topoisomerase II-mediated DNA breaks. Idarubicin is about 57.5 times and 25 times more active than doxorubicin and epirubicin, respectively [3]. Idarubicin produced a concentration-dependent decrease in cell growth with an IC50 value of about 0.01 μm. Idarubicin produces a concentration-dependent inhibition of DNA synthesis. |
In vivo study | The reduction of Idarubicin, which is dependent on ketoreductase, produces a higher stereoselectivity than most ketones, causing almost complete (13s)-epimerization. The high stereoselectivity of Idarubicin reduction may be due to Chiral Induction in the presence of an asymmetric center near the carbonyl group in the Idarubicin structure. |
